A substance’s solubility in water represents its saturation concentration in water at a defined temperature.

Water solubility is one of the fundamental properties required for the registration of chemical substances under the **REACH** regulation. For nanometric materials, this test is the first critical filter: it determines whether your product must follow the complex pathway for “nanomaterials” or whether it can be treated as a conventional soluble substance.

### Principle of the OECD 105 solubility test

The test consists of determining the **maximum concentration** of a substance dissolved in water under equilibrium conditions, at controlled temperature. Two methods are recommended for carrying out this test:

 The shaking method (Flask method)
- Suitable for substances with solubility below **10^-2 g/L**.
- The substance is suspended in water and shaken until equilibrium is reached.
- After filtration or centrifugation, the concentration is measured using an appropriate analytical technique (HPLC, UV-Vis, GC, etc.).

 The column elution method (Column Elution method)
- Suitable for substances with solubility below **10^-2 g/L**.
- Water saturates the substance in a column, and the eluate is analyzed to determine the concentration in solution.
