## Our FAQ

 What is the difference between sequencing a standard mAb and an ADC?
Sequencing an ADC is more complex because the grafted cytotoxic agents can mask certain enzymatic cleavage sites or alter the mass spectrum. At **FILAB**, we use specific protocols (sometimes including a deconjugation step) to "clean" the signal and ensure maximum sequence coverage of the carrier antibody.

 Can you precisely locate where the drug is attached (Drug Load Distribution)?
Yes. Through [LC-MS/MS](https://filab.fr/en/our-technical-resources/laboratory-analysis-uplc/laboratory-analysis-hplc-lcms.md) [Peptide Mapping](https://filab.fr/en/sectors-of-activity/biopharmaceutical/biotherapeutic-recombinant-protein-characterization/peptide-analysis-biopharmaceutical-services.md), we can identify peptides modified by the linker-drug and thus determine the conjugation sites (Cysteine, Lysine, or site-specific). This also allows us to corroborate the Drug-to-Antibody Ratio (DAR) calculation.

 Do you need the theoretical sequence to work?
It is preferable for sequence confirmation ([Peptide Mapping](https://filab.fr/en/sectors-of-activity/biopharmaceutical/biotherapeutic-recombinant-protein-characterization/peptide-analysis-biopharmaceutical-services.md)), as it accelerates data processing. However, thanks to our [De Novo sequencing](https://filab.fr/en/sectors-of-activity/biopharmaceutical/biotherapeutic-recombinant-protein-characterization/de-novo-protein-sequencing.md) capabilities by mass spectrometry, we can determine the amino acid sequence even without complete prior information.

 What sample volume is required?
Mass spectrometry techniques are highly sensitive. Generally, a few hundred micrograms (µg) of ADC are sufficient for a complete sequencing and mapping study. Contact our team to validate the quantity based on your concentration.
