What is a MALDI-TOF?

Maldi TOF (Matrix-Assisted Laser Desorption/Ionization Time-of-Flight) analysis is an analytical technique used primarily in the fields of biochemistry, molecular biology and proteomics. Maldi TOF analysis uses a laser to ionise proteins, peptides, oligonucleotides, polysaccharides and other molecules from a sample placed on a special type of target plate coated with a matrix material. The resulting ions are then accelerated in a time-of-flight tube and analysed to obtain mass spectra that can be used to identify the molecules present in the sample. Maldi TOF analysis is also often used as part of the protein identification process for targeted proteomics experiments, where it can be combined with other techniques.

Maldi/TOF mass spectroscopy involves placing a sample on a special type of target plate, coated with an organic molecule called a matrix. When the sample is exposed to a laser pulse, the matrix absorbs the energy and vaporises the molecules in the sample, releasing them as ions which are then accelerated in a time-of-flight tube. By measuring the time taken for these ions to reach the detector at the other end of the tube and correlating this with their mass, it is possible to identify the types of molecules present in the original sample. This information can be used for applications such as protein identification or targeted proteomics experiments.

Maldi/TOF analysis offers many advantages over traditional molecular analysis methods. It is highly accurate and can provide detailed information on molecules such as molecular weight, structure and abundance. In addition, it requires minimal sample preparation, can analyse several samples simultaneously and allows the analysis of non-volatile or heat sensitive materials. Maldi/TOF mass spectroscopy is also much faster than other methods, making it ideal for high-throughput applications.
