Sequencing an ADC is more complex because the grafted cytotoxic agents can mask certain enzymatic cleavage sites or alter the mass spectrum. At FILAB, we use specific protocols (sometimes including a deconjugation step) to "clean" the signal and ensure maximum sequence coverage of the carrier antibody.
Yes. Through LC-MS/MS Peptide Mapping, we can identify peptides modified by the linker-drug and thus determine the conjugation sites (Cysteine, Lysine, or site-specific). This also allows us to corroborate the Drug-to-Antibody Ratio (DAR) calculation.
It is preferable for sequence confirmation (Peptide Mapping), as it accelerates data processing. However, thanks to our De Novo sequencing capabilities by mass spectrometry, we can determine the amino acid sequence even without complete prior information.
Mass spectrometry techniques are highly sensitive. Generally, a few hundred micrograms (µg) of ADC are sufficient for a complete sequencing and mapping study. Contact our team to validate the quantity based on your concentration.
